Off two consecutive victories, Barber Road made his stakes debut in the Lively Shively Stakes at Churchill Downs on Nov. 27, 2021. He chased gamely in the stretch, but ended up second, beaten a half-length by Tejano Twist. Barber Road followed that performance with two more runner-up finishes in the Smarty Jones Stakes at Oaklawn Park on Jan. 1, 2022, and the Grade 3 Southwest Stakes on Jan. 29. Both performances earned him four points each on the road to the Kentucky Derby Presented by Woodford Reserve.

Next out, Barber Road was the 6.50-1 third choice in the Grade 2 Rebel Stakes at Oaklawn on Feb. 26. He rallied from ninth and closed to finish third, beaten by a half-length. That finish earned him 10 more Derby qualifying points. He picked up 40 more points in his final prep, the Apr. 2 Arkansas Derby, when he closed for second. In that performance, he ensured himself a spot in the starting gate on the first Saturday in May.
After rating in last in the early stages of the Derby, Barber Road made a strong, wide move in the stretch and crossed the wire sixth, beaten 4 3/4 lengths by upset winner Rich Strike.
